The MGISEQ-2000RS FCL sequencing kit features high throughput and low cost per Gb; however, in some situations users don't have enough samples to fill all the lanes, resulting in a much higher run cost. To provide a flexible package, we've developed the MGISEQ-2000RS FCS Sequencing Reagent Kit, which has lower output to make it easier to fully saturate the lanes, while also increasing run speed and flexibility to allow for different combinations of low and medium throughput sequencing. Run more, run faster!


There’s a new addition to the MGISEQ-2000RS series – a faster and more flexible MGISEQ-2000RS high-throughput rapid sequencing reagent kit.Various sample and library types tested by early access users include: tissue, FFPE, cfDNA, gDNA, multiplex PCR, hybridization products, tumor standards of different mutation frequencies, plasma free DNA capture libraries, ctDNA capture libraries. Overall both data output and quality were well above expectations.

We’ve been using PE100 of on the mgiseq-2000 platform, the PE150 of FCS can be especially suitable for some sample mutations that must be detected with a longer read length while also ensuring the same delivery time. The overall quality of the data is quite good, and the amount of data is more flexible, fully meeting the testing needs of our clinical samples.

As a versatile high-throughput gene sequencer, MGISEQ-2000 can be widely used for various applications such as WGS, WES, RNA, WGBS and Panel. However with the increasing variety of users and applications, many companies and laboratories have a strong demand for reagents with smaller data volumes, and higher run flexibility. To this end, MGI has developed a high-throughput rapid sequencing reagent kit for the comprehensive sequencer MGISEQ-2000. The reagent kit has undergone several rounds of internal testing, as well as data quality and sequencing speed optimizations. It has shown excellent performance in key areas such as data accuracy, mutation detection rate, and stability. It was also praised by our early test users. After a long process of trial and error, the company has officially launched a high-throughput rapid sequencing reagent kit.


MGISEQ-2000RS high-throughput rapid sequencing reagent kit uses the FCS slide type, which has a smaller throughput than the FCL slide. At the same time, there are also adjustments to some key components of the sequencing reagent such as reducing the optic capture area, which greatly shortens the sequencing cycle.


In the near future, MGI will also release the FCS exclusive platform MGISEQ-2000RS FAST, a faster, more flexible, and economical dual-slide mid-throughput sequencer, enabling more small and medium-sized enterprises and laboratories to be able to develop their own sequencing capabilities.

I.The remarkable features of FCS reagent


Faster Seuqencing Speed

Under the same read length, the FCS slides are 50% faster than the FCL slides. For running PE100, the FCS slide only took about 26 hours. For PE150 it takes only about 37 hours.


No Wasted Lanes

The FCS single-slide is only 550M reads, making it much easier to saturate the all the lanes with less number of samples, eliminating waste and lowering the total operational cost of a single run.


High Data Quality

More speed and the same high quality data. FCS data quality is equivalent to that of FCL.


Cross-platform compatibility

In addition to being compatibile with the existing MGISEQ-2000RS, the FCS exclusive model MGISEQ-2000RS FAST will also be released soon.

II. Test Performance

We tested the performance of the FCS reagent kit for PE150 sequencing on four MGISEQ-2000RS sequencers. This test had a total of 12 slides or 24 lanes, including 16 standard lanes of the internal standard library, as well as various application types such as tumor detection, WGS, and WES. The reults are as follows:

The data showed that the FCS PE150 sequencing reagent kit’s performance on the internal standard library were within expectations in terms of data output, data quality, and resolution.


At the same time, we also tested the performance of the FCS reagent kit for PE100 sequencing.The test was carried out on two MGISEQ-2000RS sequencers, which were divided into five slides, or 10 lanes, including 9 internal standard lanes, and application types including tumor detection. The results are as follows:

The data showed that the FCS PE100 sequencing reagent kit’s performance on the internal standard library were within expectations in terms of data volume, data quality and resolution.

The results showed that FCS was slightly better than FCL in the detection of high-frequency variants (>3%), and the detection rate of all samples reached 99%. The detection results for the intermediate frequency variants (1%-3%) were within expectations.


IV. Comprehensive evaluation
